  Abstract Understanding the genetic mechanisms of speciation and basis of species differences is among the most important challenges in evolutionary biology. Two questions of particular interest are what roles divergent selection and chromosomal differentiation play in these processes. A number of recently proposed theories argue that chromosomal rearrangements can facilitate the development and maintenance of reproductive isolation and species differences by suppressing recombination within rearranged regions. Reduced recombination permits the accumulation of alleles contributing to isolation and adaptive differentiation and protects existing differences from the homogenizing effects of introgression between incipient species. Here, we examine patterns of genetic diversity and divergence in rearranged versus collinear regions in two widespread, extensively hybridizing sunflower species, Helianthus annuus and Helianthus petiolaris, using sequence data from 77 loci distributed throughout the genomes of the two species. We find weak evidence for increased genetic divergence near chromosomal break points but not within rearranged regions overall. We find no evidence for increased rates of adaptive divergence on rearranged chromosomes; in fact, collinear chromosomes show a far greater excess of fixed amino acid differences between the two species. A comparison with a third sunflower species indicates that much of the nonsynonymous divergence between H. annuus and H. petiolaris probably occurred during or soon after their formation. Our results suggest a limited role for chromosomal rearrangements in genetic divergence, but they do document substantial adaptive divergence and provide further evidence of how species integrity and genetic identity can be maintained at many loci in the face of extensive hybridization and gene flow.

  Abstract Ants have evolved venoms rich in peptides and proteins used for predation, defense, and communication. However, they remain extremely understudied due to the minimal amount of venom secreted by each ant. The present study investigated the differences in the proteome and peptidome of the venom from the bullet ant, Paraponera clavata. Venom samples were collected from a single colony either by manual venom gland dissection or by electrical stimulation and were compared using proteomic methods. Venom proteins were separated by 2D-PAGE and identified by nanoLC-ESI-QTOF MS/MS. Venom peptides were initially separated using C18 reversed-phase high-performance liquid chromatography, then analyzed by MALDI-TOF MS. The proteomic analysis revealed numerous proteins that could be assigned a biological function (total 94), mainly as toxins, or roles in cell regulation and transport. This investigation found that ca. 73% of the proteins were common to venoms collected by the two methods. The peptidomic analysis revealed a large number of peptides (total 309) but with <20% shared by the two collection methods. There was also a marked difference between venoms obtained by venom gland dissection from different ant colonies. These findings demonstrate the rich composition and variability of P. clavata venom.

  Abstract P>Cross-species analyses of plant functional traits have shed light on factors contributing to differences in performance and distribution, but to date most studies have focused on either leaves or stems. We extend these tissue-specific analyses of functional strategy towards a whole-plant approach by integrating data on functional traits for 13 448 leaves and wood tissues from 4672 trees representing 668 species of Neotropical trees. Strong correlations amongst traits previously defined as the leaf economics spectrum reflect a tradeoff between investments in productive leaves with rapid turnover vs. costly physical leaf structure with a long revenue stream. A second axis of variation, the 'stem economics spectrum', defines a similar tradeoff at the stem level: dense wood vs. high wood water content and thick bark. Most importantly, these two axes are orthogonal, suggesting that tradeoffs operate independently at the leaf and at the stem levels. By simplifying the multivariate ecological strategies of tropical trees into positions along these two spectra, our results provide a basis to improve global vegetation models predicting responses of tropical forests to global change.

  Abstract We assessed the extent of recent environmental changes on leaf morphological (stomatal density, stomatal surface, leaf mass per unit area) and physiological traits (carbon isotope composition, delta(13)C(leaf), and discrimination, Delta(13)C(leaf), oxygen isotope composition, delta(18)O(leaf)) of two tropical rainforest species (Dicorynia guianensis; Humiria balsamifera) that are abundant in the Guiana shield (Northern Amazonia). Leaf samples were collected in different international herbariums to cover a 200 year time-period (1790-2004) and the whole Guiana shield. Using models describing carbon and oxygen isotope fractionations during photosynthesis, different scenarios of change in intercellular CO(2) concentrations inside the leaf (C(i)), stomatal conductance (g), and photosynthesis (A) were tested in order to understand leaf physiological response to increasing air CO(2) concentrations (C(a)). Our results confirmed that both species displayed physiological response to changing C(a). For both species, we observed a decrease of about 1.7% in delta(13)C(leaf) since 1950, without significant change in Delta(13)C(leaf) and leaf morphological traits. Furthermore, there was no clear change in delta(18)O(leaf) for Humiria over this period. Our simulation approach revealed that an increase in A, rather than a decrease in g, explained the observed trends for these tropical rainforest species, allowing them to maintain a constant ratio of C(i)/C(a).

  Abstract Functional traits provide insight into a variety of ecological questions, yet the optimal sampling method to estimate the community-level distribution of plant functional trait values remains a subject of debate, especially in species-rich forests. We present a simulation analysis of the trait distribution of a set of nine completely sampled permanent plots in the lowland rain forests of French Guiana. Increased sampling intensity consistently improved accuracy in estimating community-weighted means and variances of functional trait values, whereas there was substantial variation among functional traits and minor differences among sampling strategies. Thus, investment in intensified sampling yields a greater improvement in the accuracy of estimation than does an equivalent investment in sampling design complication. Notably, 'taxon-free' strategies frequently had greater accuracy than did abundance-based strategies, which had the additional cost of requiring botanical surveys. We conclude that there is no substitute for extensive field sampling to accurately characterize the distribution of functional trait values in species-rich forests. © 2015 British Ecological Society.

  Abstract The biological activity of extracts from the leaves, bark and roots of Muellera frutescens, an Amazonian ichtyotoxic plant, were evaluated to find new environmentally safe insecticides. The n-hexane extracts of bark, leaf, and root showed a strong toxic activity against Aedes aegypti mosquito larvae. Bioguided fractionation of the bark extract led to the isolation of seven isoflavonoids (12a-hydroxyelliptone, elliptone, (-)-variabilin, rotenone, rotenolone, tephrosin and deguelin). Rotenone and deguelin are responsible for the larvicidal activity of the plant. M. frutescens leaves contain up to 0.6%, w/w, deguelin. These results justify the traditional ichtyotoxic use of M. frutescens. The leaves contain a relatively high proportion of deguelin and, therefore, can be considered as a renewable source of this environmentally friendly insecticidal isoflavonoid.

  Abstract Theory suggests that spatial structuring should select for intermediate levels of virulence in parasites, but empirical tests are rare and have never been conducted with castration (sterilizing) parasites. To test this theory in a natural landscape, we construct a spatially explicit model of the symbiosis between the ant-plant Cordia nodosa and its two, protecting ant symbionts, Allomerus and Azteca. Allomerus is also a castration parasite, preventing fruiting to increase colony fecundity. Limiting the dispersal of Allomerus and host plant selects for intermediate castration virulence. Increasing the frequency of the mutualist, Azteca, selects for higher castration virulence in Allomerus, because seeds from Azteca-inhabited plants are a public good that Allomerus exploits. These results are consistent with field observations and, to our knowledge, provide the first empirical evidence supporting the hypothesis that spatial structure can reduce castration virulence and the first such evidence in a natural landscape for either mortality or castration virulence.

  Abstract Aim: To examine variation in the phylogenetic diversity (PD) of tree communities across geographical and environmental gradients in Amazonia. Location: Two hundred and eighty-three c. 1 ha forest inventory plots from across Amazonia. Methods: We evaluated PD as the total phylogenetic branch length across species in each plot (PDss), the mean pairwise phylogenetic distance between species (MPD), the mean nearest taxon distance (MNTD) and their equivalents standardized for species richness (ses.PDss, ses.MPD, ses.MNTD). We compared PD of tree communities growing (1) on substrates of varying geological age; and (2) in environments with varying ecophysiological barriers to growth and survival. Results: PDss is strongly positively correlated with species richness (SR), whereas MNTD has a negative correlation. Communities on geologically young- and intermediate-aged substrates (western and central Amazonia respectively) have the highest SR, and therefore the highest PDss and the lowest MNTD. We find that the youngest and oldest substrates (the latter on the Brazilian and Guiana Shields) have the highest ses.PDss and ses.MNTD. MPD and ses.MPD are strongly correlated with how evenly taxa are distributed among the three principal angiosperm clades and are both highest in western Amazonia. Meanwhile, seasonally dry tropical forest (SDTF) and forests on white sands have low PD, as evaluated by any metric. Main conclusions: High ses.PDss and ses.MNTD reflect greater lineage diversity in communities. We suggest that high ses.PDss and ses.MNTD in western Amazonia results from its favourable, easy-to-colonize environment, whereas high values in the Brazilian and Guianan Shields may be due to accumulation of lineages over a longer period of time. White-sand forests and SDTF are dominated by close relatives from fewer lineages, perhaps reflecting ecophysiological barriers that are difficult to surmount evolutionarily. Because MPD and ses.MPD do not reflect lineage diversity per se, we suggest that PDss, ses.PDss and ses.MNTD may be the most useful diversity metrics for setting large-scale conservation priorities. © 2015 John Wiley & Sons Ltd.

  Abstract Coarse woody debris (CWD, dead wood sections ≥10 cm diameter) represents a large store of carbon in tropical forests; however, estimates of the flux of carbon from CWD in these forests remain poorly constrained. The objective of this study was to resolve the dry/wet season response of respiration in CWD (Rcwd), and investigate the importance of biotic and abiotic factors for predicting the seasonal change of Rcwd at the ecosystem level. This study presents a 4-month time series of Rcwd measurements conducted on 42 dead trees (26 species) at the Paracou Research Station in French Guiana. Rcwd measurements were repeated 13 times on each CWD sample from July to November 2011, spanning the transition from wet to dry season, and then from dry season to the following wet season. Seasonal drought caused monthly Rcwd to drop by 20.5 ± 5.1% over the wet-dry transition. Changes in woody tissue moisture content explained 41.9% of the measured seasonal variability in Rcwd, but 60% of the seasonal variability in mean forest Rcwd rates could be modelled using surface soil water content. We estimate that Rcwd is approximately 5% of annual ecosystem respiration (Reco) and that seasonal variations in Rcwd contribute appreciably to seasonal variations of Reco, and should be included in functional models simulating the response of tropical rainforest ecosystems to current and future climate. © 2013 Springer Science+Business Media New York.

  Abstract In order to progress in the understanding of mechanical stress generation, the mesoporosity of the cell wall and its changes during maturation of poplar (Populus deltoides × P. nigra) tension wood (TW) and opposite wood (OW) were measured by nitrogen adsorption-desorption. Variations in the thickness of the gelatinous layer (G-layer) were also measured to clarify whether the mesoporosity change simultaneously with the deposition of the G-layer in TW. Results show that mesoporous structures of TW and OW were very similar in early development stages before the deposition of G-layers. With the formation of the S2 layer in OW and the G-layer in TW, the mesopore volume decreased steeply before lignification. However, in TW only, the decrease in mesopore volume occurred together with the pore shape change and a progressive increase in pore size. The different patterns observed in TW revealed that pores from G-layers appear with a different shape compared to those of the compound middle lamella, and their size increases during the maturation process until stabilising in mature wood. This observation strongly supports the hypothesis of the swelling of the G-layer matrix during maturation as the origin of maturation stress in poplar tension wood.
