Abstract: Animal venom peptides are currently being developed as novel drugs and bioinsecticides. Because ants use venoms for defense and predation, venomous ants represent an untapped source of potential bioactive toxins. This study compared the protein and peptide components of the poneroid ants Neoponera commutata, Neoponera apicalis, and Odontomachus hastatus and the formicoid ants Ectatomma tuberculatum, Ectatomma brunneum, and Myrmecia gulosa. 1D and 2D PAGE revealed venom proteins in the mass range <10 to >250 kDa. NanoLC-ESI-QTOF MS/MS analysis of tryptic peptides revealed the presence of common venom proteins and also many undescribed proteins. RP-HPLC separation followed by MALDI-TOF MS of the venom peptides also revealed considerable heterogeneity. It was found that the venoms contained between 144 and 1032 peptides with 5-95% of peptides in the ranges 1-4 and 1-8 kDa for poneroid and formicoid ants, respectively. By employing the reducing MALDI matrix 1,5-diaminonapthalene, up to 28 disulfide-bonded peptides were also identified in each of the venoms. In particular, the mass range of peptides from poneroid ants is lower than peptides from other venoms, indicating possible novel structures and pharmacologies. These results indicate that ant venoms represent an enormous, untapped source of novel therapeutic and bioinsecticide leads. © 2016 American Chemical Society.

Abstract: Ethnopharmacological relevance Psidium acutangulum Mart. ex DC is a small tree used by the Wayana Amerindians from the Upper-Maroni in French Guiana for the treatment of malaria. Aim of the study In a previous study, we highlighted the in vitro antiplasmodial, antioxidant and anti-inflammatory potential of the traditional decoction of P. acutangulum aerial parts. Our goal was then to investigate on the origin of the biological activity of the traditional remedy, and eventually characterize active constituents. Materials and methods Liquid-liquid extractions were performed on the decoction, and the antiplasmodial activity evaluated against chloroquine-resistant FcB1 ([3H]-hypoxanthine bioassay) and 7G8 (pLDH bioassay) P. falciparum strains, and on a chloroquine sensitive NF54 ([3H]-hypoxanthine bioassay) P. falciparum strain. The ethyl acetate fraction (D) was active and underwent bioguided fractionation. All the isolated compounds were tested on P. falciparum FcB1 strain. In vitro anti-inflammatory activity (IL-1β, IL-6, IL-8, TNFα) of the ethyl acetate fraction and of an anti-Plasmodium active compound, was concurrently assessed on LPS-stimulated human PBMC and NO secretion inhibition was measured on LPS stimulated RAW murine macrophages. Cytotoxicity of the fractions and pure compounds was measured on VERO cells, L6 mammalian cells, PBMCs, and RAW cells. Results Fractionation of the ethyl acetate soluble fraction (IC50 ranging from 3.4 to <1 μg/mL depending on the parasite strain) led to the isolation of six pure compounds: catechin and five glycosylated quercetin derivatives. These compounds have never been isolated from this plant species. Two of these compounds (wayanin and guaijaverin) were found to be moderately active against P. falciparum FcB1 in vitro (IC50 5.5 and 6.9 μM respectively). We proposed the name wayanin during public meetings organized in June 2015 in the Upper-Maroni villages, in homage to the medicinal knowledge of the Wayana population. At 50 μg/mL, the ethyl acetate fraction (D) significantly inhibited IL-1β secretion (-46%) and NO production (-21%), as previously observed for the decoction. The effects of D and guiajaverin (4) on the secretion of other cytokines or NO production were not significant. Conclusions The confirmed antiplasmodial activity of the ethyl acetate soluble fraction of the decoction and of the isolated compounds support the previous results obtained on the P. acutangulum decoction. The antiplasmodial activity might be due to a mixture of moderately active non-toxic flavonoids. The anti-inflammatory activities were less marked for ethyl acetate fraction (D) than for the decoction. © 2016 Elsevier Ireland Ltd. All rights reserved.

Abstract: In this study, the corrosion of S355 carbon steel was monitored for 50 days in equatorial environment. The experiments were conducted under three complementary approaches in a natural environment: (i) bacterial diversity was assessed with Miseq sequencing, (ii) observations of the surface and identifications of deposit compounds were realised with scanning electron microscopy (SEM), Raman spectrometry and EDX analyses, (iii) electrochemical measurements were used to calculate corrosion rate and to identify the phenomena which control corrosion. The results showed that the bacterial population had changed over immersion time from a dominance of β-proteobacteria to α-proteobacteria. This evolution decreased charge transfer resistance at the metal/deposit from 41.6 to 20.8 Ω/cm2 in 50 days, caused an increase in the corrosion rate by a factor of 2, from 0.13 to 0.27 mm/year. Surface analyses showed a progressive appearance of MIC markers in the deposit such as iron sulphide and manganese oxides that were detected after 30 days of immersion in the natural environment. In conclusion, this study describes in detail, the first stage of MIC activity on carbon steel surface in an equatorial brackish water, under aerobic conditions. © 2016 Elsevier Ltd.

Abstract: Aims One critical challenge for plants is to maintain an adequate nutrient supply under fluctuating environmental conditions. This is particularly true for epiphytic species that have limited or no access to the pedosphere and often live in harsh climates. Bromeliads have evolved key innovations such as epiphytism, water-absorbing leaf trichomes, tank habit and Crassulacean acid metabolism (CAM) photosynthesis that enable them to survive under various environmental conditions. Bromeliads encompass diverse ecological types that live on different substrates (they can be terrestrial, epilithic or epiphytic) and vary in their ability to retain water (they can be tank-forming or tankless) and photosynthetic pathway (i.e. C3 or CAM). In this review, we outline the nutritional modes and specializations that enable bromeliads to thrive in a wide range of nutrient-poor (mostly nitrogen-depleted) environments. Important Findings Bromeliads have evolved a great diversity of morphologies and functional adaptations leading to the existence of numerous nutritional modes. Focusing on species that have absorptive foliar trichomes, we review evidence that bromeliads have evolved multi-faceted nutritional strategies to respond to fluctuations in the supply of natural nitrogen (N). These plants have developed mutualistic associations with many different and functionally diverse terrestrial and aquatic microorganisms and metazoans that contribute substantially to their mineral nutrition and, thus, their fitness and survival. Bacterial and fungal microbiota-assisted N provisioning, protocarnivory, digestive mutualisms and myrmecotrophic pathways are the main strategies used by bromeliads to acquire nitrogen. The combination of different nutritional pathways in bromeliads represents an important adaptation enabling them to exploit nutrient-poor habitats. Nonetheless, as has been shown for several other vascular plants, multiple partners are involved in nutrient acquisition indicating that there have been convergent adaptations to nutrient scarcity. Finally, we point out some gaps in the current knowledge of bromeliad nutrition that offer fascinating research opportunities. © The Author 2015. Abstract: Ecosystems are being stressed by climate change, but few studies have tested food web responses to changes in precipitation patterns and the consequences to ecosystem function. Fewer still have considered whether results from one geographic region can be applied to other regions, given the degree of community change over large biogeographic gradients. We assembled, in one field site, three types of macroinvertebrate communities within water-filled bromeliads. Two represented food webs containing both a fast filter feeder–microbial and slow detritivore energy channels found in Costa Rica and Puerto Rico, and one represented the structurally simpler food webs in French Guiana, which only contained the fast filter feeder–microbial channel. We manipulated the amount and distribution of rain entering bromeliads and examined how food web structure mediated ecosystem responses to changes in the quantity and temporal distribution of precipitation. Food web structure affected the survival of functional groups in general and ecosystem functions such as decomposition and the production of fine particulate organic matter. Ecosystem processes were more affected by decreased precipitation than were the abundance of micro-organisms and metazoans. In our experiments, the sensitivity of the ecosystem to precipitation change was primarily revealed in the food web dominated by the single filter feeder–microbial channel because other top-down and bottom-up processes were weak or absent. Our results show stronger effects of food web structure than precipitation change per se on the functioning of bromeliad ecosystems. Consequently, we predict that ecosystem function in bromeliads throughout the Americas will be more sensitive to changes in the distribution of species, rather than to the direct effects caused by changes in precipitation. © 2016 The Authors.
