  Abstract This study is part of the activities carried out to monitor tow-impact logging operations in humid tropical forests. The sustainability of these activities is highly dependent on the scale of logging damage, and managers therefore need to carry out effective checks on the quality of forestry work. In this study, remote sensing was used to track the extension of logging operations and to assess damage, through images acquired during and after logging in each parcel. Analysis of the high-resolution images (10 m) clearly identifies logged-over clearings and some skidding tracks, allowing assessments of gaps opening up in the canopy. Several parcels were monitored in this French Guiana study. A simple diagnostic procedure was established for each parcel to assess logging intensity and identify new logging tracks and timber losses. The remote sensing toot thus helps to make more accurate assessments of logging quality and to draw relevant conclusions for the logging company. The study has demonstrated the usefulness of remote sensing to identify damage after logging. The quality of restitution of the damage was validated by means of a database on the logging terrain and through field missions. This post-logging inspection tool can now be automated for fully operational use by forest managers.

  Abstract Tropical forests harbor the greatest terrestrial biodiversity and provide various ecosystem services. The increase of human activities on these forests, among which logging, makes the conservation of biodiversity and associated services strongly dependent on the sustainability of these activities. However the indicators commonly used to assess the impact of forest exploitation, namely species richness and biomass, provide a limited understanding of their sustainability. Here, we assessed the sustainability of common forest exploitation in the Guiana Shield studying the recovery of two ecosystem services i.e. carbon storage and wood stock, and an ecosystem function i.e. seed dispersal by animals. Specifically, we compared total and commercial biomass, as well as functional composition in seed size of animal-dispersed species in replicated forest plots before and 27 years after exploitation. Species richness is also studied to allow comparison. While species richness was not affected by forest exploitation, total and commercial biomass as well as seed size of animal-dispersed species decreased 27 years after exploitation, similarly to forests affected by hunting. These results show that ecosystem services and function likely did not recover even at the lowest intensity of forest exploitation studied, questioning the sustainability of the most common rotation-cycle duration applied in the tropics.

  Abstract Bioassay-guided fractionation of the ethyl acetate bark extract of Rheedia acuminata led to the isolation of the new compound 1,5,6-trihydroxy-3-methoxy-7-geranyl-xanthone (1), together with four known compounds 2-5. These compounds were tested in vitro for their antiplasmodial activity on a chloroquine-resistant strain of Plasmodium falciparum (FcB1) and for their cytotoxicity against the human diploid embryonic lung cell line MRC-5.

  Abstract Previous studies indicate that some populations of the little fire ant, Wasmannia auropunctata, display an unusual reproduction system polymorphism. Although some populations have a classical haplodiploid reproduction system, in other populations queens are produced by thelytokous parthenogenesis, males are produced by a male clonality system and workers are produced sexually. An atypical genetic caste determination system was also suggested. However, these conclusions were indirectly inferred from genetic studies on field population samples. Here we set up experimental laboratory nests that allow the control of the parental relationships between individuals. The queens heading those nests originated from either putatively clonal or sexual populations. We characterized the male, queen and worker offspring they produced at 12 microsatellite loci. Our results unambiguously confirm the unique reproduction system polymorphism mentioned above and that male clonality is strictly associated with thelytokous parthenogenesis. We also observed direct evidence of the rare production of sexual gynes and arrhenotokous males in clonal populations. Finally, we obtained evidence of a genetic basis for caste determination. The evolutionary significance of the reproduction system polymorphism and genetic caste determination as well as future research opportunities are discussed.   Abstract In order to face environmental constraints, trees are able to re-orient their axes by controlling the stress level in the newly formed wood layers. Angiosperms and gymnosperms evolved into two distinct mechanisms: the former produce a wood with large tension pre-stress on the upper side of the tilted axis, while the latter produce a wood with large compression pre-stress on the lower side. In both cases, the difference between this stress level and that of the opposite side, in light tension, generates the bending of the axis. However, light values of compression were sometimes measured in the opposite side of angiosperms. By analysing old data on chestnut and mani and new data on poplar, this study shows that these values were not measurement artefacts. This reveals that generating light compression stress in opposite wood contributes to improve the performance of the re-orientation mechanism.

  Abstract Genetic variation for intrinsic water use efficiency (W-i) and related traits was estimated in a full-sib family of Quercus robur L. over 3 years. The genetic linkage map available for this F1 family was used to locate quantitative trait loci (QTL) for W-i, as estimated by leaf carbon stable isotope composition (delta C-13) or the ratio of net CO2 assimilation rate (A) to stomatal conductance to water vapour (g(w)) and related leaf traits. Gas exchange measurements were used to standardize estimates of A and g(w) and to model the sensitivity of gw to leaf-to-air vapour pressure deficit (sg(VPD)). delta C-13 varied by more than 3% among the siblings, which is equivalent to 40% variation of W-i. Most of the studied traits exhibited high clonal mean repeat-abilities (> 50%; proportion of clonal mean variability in global variance). Repeatabilities for delta C-13, leaf mass per area (LMA) and leaf nitrogen content were higher than 70%. For delta C-13, ten QTLs were detected, one of which was detected repeatedly for all 3 years and consistently explained more than 20% of measured variance. Four genomic regions were found in which co-localizing traits linked variation in W-i to variations in leaf chlorophyll and nitrogen content, LMA and sg(VPD). A positive correlation using clonal means between delta C-13 and A/g(w), as well as a co-localisation of QTL detected for both traits, can be seen as validation of the theoretical model linking the genetic architecture of these two traits.

  Abstract Aim of the study: Our objective was to assess whether it could be contemplated to recommend Quassia amara young leaf tea for treatment against malaria. and if yes. set up a standard protocol for preparing the herbal tea. Materials and methods: The leaf tea was extracted with methylene chloride and the organic extract was fractionated with HPLC. Pure compounds were characterized and their in vitro cytotoxicity and antiplasmodial activity was determined. Results and discussion: We discovered that antimalarial Quassia amara young leaf tea contains several quassinoids: simalikalactone D (SkD. 1), picrasin B (2). picrasin H (3), neoquassin (4), quassin (5), picrasin 1(6) and picrasin J (7). These last two compounds are new. In addition. our experiments demonstrate that both biological activity and cytotoxicity of the remedy may be attributed solely to the presence of SkD. Conclusion: In conclusion, this preparation Should not be recommended for treatment of malaria until a clinical Study in humans is performed with SkD. (C) 2009 Elsevier Ireland Ltd.   Abstract Pedunculate oak and sessile oak are two sympatric inter-fertile species that exhibit leaf morphological differences. We aimed to detect quantitative trait loci (QTLs) of these traits in order to locate genomic regions involved in species differentiation. A total of 15 leaf morphological traits were assessed in a mixed forest stand composed of Quercus petraea and Q. robur and in a full-sib pedigree of Q. robur. The progeny of the full-sib family were vegetatively propagated in two successive experiments comprising 174 and 216 sibs, and assessments were made on two leaves collected on each of the 1080 and 1530 cuttings corresponding to the two experiments. Traits that exhibited strong species differences in the mixed stand tended also to have higher repeatability values in the mapping population, thus indicating higher genetic control. A genetic map was constructed for QTL detection. Composite interval mapping with the one QTL model was used for QTL detection. From one to three QTLs were detected for 13 traits. In-depth analysis of the QTLs, controlling the five morphological traits that exhibited the highest interspecific differences in the mixed stand, indicated that they were distributed on six linkage groups, with two clusters comprising QTLs of at least two discriminant traits. These results were reinforced when error 1 for QTL detection was set at 5% at the chromosome level, as up to nine clusters could be identified. In conclusion, traits involved in interspecific differentiation of oaks are under polygenic control and widespread in clusters across the genome.

  Abstract Oak chloroplast DNA (cpDNA) variation was studied in a grid-based inventory in western Central Europe, including Belgium, The Netherlands, Luxembourg, Germany, the Czech Republic, and the northern parts of Upper and Lower Austria. A total of 2155 trees representing 426 populations of Quercus robur L. and Q. petraea (Matt.) Liebl. were screened for polymorphism in up to four PCR-amplified cpDNA fragments. Eleven haplotypes belonging to four lineages were detected; these lineages were formerly restricted to glacial refugia in the Iberian Peninsula, the Apennine Peninsula and the Balkan Peninsula. The haplotypes originating from the Apennines are particularly well represented in the study region, but there is also a significant contribution from the other refugia, which explain the high overall level of cpDNA diversity. The strong human impact in western Central Europe during the past centuries, which has resulted in the clearance of most forests, was followed by reforestation, sometimes involving seed transfers. Despite this strong human impact, broad geographic patterns of lineages and haplotypes could still be detected. To evaluate further the consequences of the former human activities on the present day oak cpDNA genetic structure, four regions where increasingly strong human impact was anticipated (ranging from hilly regions in southern Germany to roadsides plantations in The Netherlands) were selected. There, a comparison of the levels of intrapopulation cpDNA diversity and spatial structuring was made. Over the whole area, within stand diversity was significantly higher in Q. robur than in Q. petraea (h(S) = 0.24 vs. 0.16). Since total diversity is identical for both species, this results in a significantly lower level of fixation for Q. robur than for Q. petraea (G(ST) = 0.68 vs. 0.79). The analyses also reveal a decrease of fixation with increasing human impact on oak populations. The Dutch roadside plantations (Q. robur) exhibit a very low level of fixation (G(ST) = 0.28) as compared to Q. petraea in southern Germany (G(ST) = 0.91). The significance of the spatial genetic structure was tested using geostatistical methods. For the complete data set, a strong spatial genetic structure is confirmed, with higher than average genetic similarities between populations distant from up to 270 km, whereas there is no spatial structure in the roadside plantations in The Netherlands or in the northern German lowlands. These results should help to differentiate introduced from autochthonous populations, and provide a framework for the identification of the geographic origin of seed lots.   Abstract Ethnopharmacological relevance:
Field investigations highlighted the use of Psidium acutangulum Mart. ex DC (syn. P. persoonii McVaugh), a small tree used by the Wayana Amerindians in Twenke–Taluhwen and Antecume–Pata, French Guiana, for the treatment of malaria, and administered either orally in the form of a decoction or applied externally over the whole body. This use appears limited to the Wayana cultural group in French Guiana and has never been reported anywhere else. Our goal was to evaluate the antimalarial and anti-inflammatory activities of a P. acutangulum decoction to explain the good reputation of this remedy.
Materials and methods:
Interviews with the Wayana inhabitants of Twenke–Taluhwen and Antecume–Pata were conducted within the TRAMAZ project according to the TRAMIL methodology, which is based on a quantitative and qualitative analysis of medicinal plant uses. A decoction of dried aerial parts of P. acutangulum was prepared in consistency with the Wayana recipe. In vitro antiplasmodial assays were performed on chloroquine-resistant FcB1 ([3H]-hypoxanthine bioassay) and 7G8 (pLDH bioassay) P. falciparum strains and on chloroquine sensitive NF54 ([3H]-hypoxanthine bioassay) P. falciparum strain. In vitro anti-inflammatory activity (IL-1β, IL-6, IL-8, TNFα) was evaluated on LPS-stimulated human PBMC and NO secretion inhibition was measured on LPS stimulated RAW murine macrophages. Cytotoxicity of the decoction was measured on L6 mammalian cells, PBMCs, and RAW cells. A preliminary evaluation of the in vivo antimalarial activity of the decoction, administered orally twice daily, was assessed by the classical four-day suppressive test against P. berghei NK65 in mice.
Results:
The decoction displayed a good antiplasmodial activity in vitro against the three tested strains, regardless to the bioassay used, with IC50 values of 3.3 µg/mL and 10.3 µg/mL against P. falciparum FcB1 and NF54, respectively and 19.0 µg/mL against P. falciparum 7G8. It also exhibited significant anti-inflammatory activity in vitro in a dose dependent manner. At a concentration of 50 µg/mL, the decoction inhibited the secretion of the following pro-inflammatory cytokines: TNFα (−18%), IL-1β (−58%), IL-6 (−32%), IL-8 (−21%). It also exhibited a mild NO secretion inhibition (−13%) at the same concentration. The decoction was non-cytotoxic against L6 cells (IC50>100 µg/mL), RAW cells and PBMC. In vivo, 150 µL of the decoction given orally twice a day (equivalent to 350 mg/kg/day of dried extract) inhibited 39.7% average parasite growth, with more than 50% of inhibition in three mice over five. The absence of response for the two remaining mice, however, induced a strong standard deviation.
Conclusions:
This study highlighted the in vitro antiplasmodial activity of the decoction of P. acutangulum aerial parts, used by Wayana Amerindians from the Upper-Maroni in French Guiana in case of malaria. Its antioxidant and anti-inflammatory potential, which may help to explain its use against this disease, was demonstrated using models of artificially stimulated cells.
