  Abstract Assessing spider diversity remains a great challenge, especially in tropical habitats where dozens of species can locally co-occur. Pitfall trapping is one of the most widely used techniques to collect spiders, but it suffers from several biases, and its accuracy likely varies with habitat complexity. In this study, we compared the efficiency of passive pitfall trapping versus active nocturnal hand collecting (\"HC) to capture low understory-dwelling spider taxonomical (morpho-species) and functional (hunting guilds) diversity along a structural gradient of habitats in French Guiana. We focused on four habitats describing a structural gradient: garden to the orchard to the forest edge to the undisturbed forest. Overall, estimated morpho-species richness and composition did not vary consistently between habitats, but abundances of ground-hunting spiders decreased significantly with increasing habitat complexity. We found habitat-dependence differences in taxonomic diversity between sampling strategies: NHC revealed higher diversity in the orchard, whereas pitfalls resulted in higher diversity in the forest. Species turnover resulted in high dissimilarity in species composition between habitats using either method. This study shows how pitfall trapping is influenced by habitat structure, rendering this sampling method incomplete for complex, tropical environments. However, pitfall traps remain a valuable component of inventories because they sample distinct assemblage of spiders. © 2020 by the authors.

  Abstract Tropical forests have exceptional floristic diversity, but their characterization remains incomplete, in part due to the resource intensity of in-situ assessments. Remote sensing technologies can provide valuable, cost-effective, large-scale insights. This study investigates the combined use of airborne LiDAR and imaging spectroscopy to map tree species at landscape scale in French Guiana. Binary classifiers were developed for each of 20 species using linear discriminant analysis (LDA), regularized discriminant analysis (RDA) and logistic regression (LR). Complementing visible and near infrared (VNIR) spectral bands with short wave infrared (SWIR) bands improved the mean average classification accuracy of the target species from 56.1% to 79.6%. Increasing the number of non-focal species decreased the success rate of target species identification. Classification performance was not significantly affected by impurity rates (confusion between assigned classes) in the non-focal class (up to 5% of bias), provided that an adequate criterion was used for adjusting threshold probability assignment. A limited number of crowns (30 crowns) in each species class was sufficient to retrieve correct labels effectively. Overall canopy area of target species was strongly correlated to their basal area over 118 ha at 1.5 ha resolution, indicating that operational application of the method is a realistic prospect (R2 = 0.75 for six major commercial tree species). © 2020 by the authors.

  Abstract A critical hurdle in ant venom proteomic investigations is the lack of databases to comprehensively and specifically identify the sequence and function of venom proteins and peptides. To resolve this, we used venom gland transcriptomics to generate a sequence database that was used to assign the tandem mass spectrometry (MS) fragmentation spectra of venom peptides and proteins to specific transcripts. This was performed alongside a shotgun liquid chromatography-mass spectrometry (LC-MS/MS) analysis of the venom to confirm that these assigned transcripts were expressed as proteins. Through the combined transcriptomic and proteomic investigation of Paraponera clavata venom, we identified four times the number of proteins previously identified using 2D-PAGE alone. In addition to this, by mining the transcriptomic data, we identified several novel peptide sequences for future pharmacological investigations, some of which conform with inhibitor cysteine knot motifs. These types of peptides have the potential to be developed into pharmaceutical or bioinsecticide peptides. © 2020 by the authors. Licensee MDPI, Basel, Switzerland.

  Abstract Near infrared (NIR) spectroscopy provides rapid and non-destructive analysis of wood properties and composition. In this study, we aimed to use NIR measurement for the prediction of teak phenolic compounds, which are chemical markers for natural durability of wood. Twenty-seven teak trees from two geographical zones (Malaysia and Ivory Coast) were used. On ground heartwood samples, the content of total phenolics and individual quinones (tectoquinone, 2-(hydroxymethyl)anthraquinone, 2-anthraquinone carboxylic acid, 1,4-naphthoquinoneand 4c,5c-dihydroxy-epiisocatalponol) were determined using high performance liquid chromatography (HPLC). Partial least squares (PLS) regression with NIR spectra on the same samples and phenolic data was used to build NIR models for phenolic contents. The PLS models for the total predicted phenolics and three quinone contents (tectoquinone, 2-(hydroxymethyl) anthraquinone, and 4¢,5¢-dihydroxy-epiisocatalponol) showed a good ratio of performance to deviation (RPD ≥ 2.5), strong coefficients of determination (r2 ≥ 0.8) and the prediction errors were consistent with the reference method. These results demonstrate that NIR spectroscopy can be reliable for the evaluation of total phenolics and individual quinones in teak heartwood wood meal. NIR spectroscopy is a promising technique for rapidly providing information on the quinone contents in teak wood and indirectly for knowing its natural durability. This finding leads to a precise, non-destructive tool for teak wood quality evaluation. © IM Publications LLP 2014.

  Abstract For highly specialized insect herbivores, plant chemical defenses are often co-opted as cues for oviposition and sequestration. In such interactions, can plants evolve novel defenses, pushing herbivores to trade off benefits of specialization with costs of coping with toxins? We tested how variation in milkweed toxins (cardenolides) impacted monarch butterfly ( Danaus plexippus ) growth, sequestration, and oviposition when consuming tropical milkweed ( Asclepias curassavica ), one of two critical host plants worldwide. The most abundant leaf toxin, highly apolar and thiazolidine ring–containing voruscharin, accounted for 40% of leaf cardenolides, negatively predicted caterpillar growth, and was not sequestered. Using whole plants and purified voruscharin, we show that monarch caterpillars convert voruscharin to calotropin and calactin in vivo, imposing a burden on growth. As shown by in vitro experiments, this conversion is facilitated by temperature and alkaline pH. We next employed toxin-target site experiments with isolated cardenolides and the monarch’s neural Na + /K + -ATPase, revealing that voruscharin is highly inhibitory compared with several standards and sequestered cardenolides. The monarch’s typical >50-fold enhanced resistance to cardenolides compared with sensitive animals was absent for voruscharin, suggesting highly specific plant defense. Finally, oviposition was greatest on intermediate cardenolide plants, supporting the notion of a trade-off between benefits and costs of sequestration for this highly specialized herbivore. There is apparently ample opportunity for continued coevolution between monarchs and milkweeds, although the diffuse nature of the interaction, due to migration and interaction with multiple milkweeds, may limit the ability of monarchs to counteradapt.

The sampled populations were selected on the basis of their contrasting time of bud burst observed in common garden experiments (provenance tests). The candidate genes were selected according to their expression profiles during the transition from quiescent to developing buds and/or their functional role in model plants. The overall nucleotide diversity was large (pi(tot) – 6.15 x 10(-3); pi(silent) – 11.2 x 10(-3)), but population differentiation was not larger than for microsatellites. No outlier single-nucleotide polymorphism (SNP), departing from neutral expectation, was found among the total of 125 SNPs. These results contrasted markedly with the significant associations that were observed between the candidate genes and bud burst in segregating populations. Quantitative trait loci (QTLs) for bud burst were identified for 13 year*site seasonal observations in a cloned mapping pedigree. Nineteen QTLs were detected, and QTLs located on linkage groups 2, 5 and 9 contributed repeatedly to more than 12% of the phenotypic variation of the trait. Eight genes were polymorphic in the two parents of the pedigree and could be mapped on the existing genetic map. Five of them located within the confidence intervals of QTLs for bud burst.   Abstract Previous studies indicate that some populations of the little fire ant, Wasmannia auropunctata, display an unusual reproduction system polymorphism. Although some populations have a classical haplodiploid reproduction system, in other populations queens are produced by thelytokous parthenogenesis, males are produced by a male clonality system and workers are produced sexually. An atypical genetic caste determination system was also suggested. However, these conclusions were indirectly inferred from genetic studies on field population samples. Here we set up experimental laboratory nests that allow the control of the parental relationships between individuals. The queens heading those nests originated from either putatively clonal or sexual populations. We characterized the male, queen and worker offspring they produced at 12 microsatellite loci. Our results unambiguously confirm the unique reproduction system polymorphism mentioned above and that male clonality is strictly associated with thelytokous parthenogenesis. We also observed direct evidence of the rare production of sexual gynes and arrhenotokous males in clonal populations. Finally, we obtained evidence of a genetic basis for caste determination. The evolutionary significance of the reproduction system polymorphism and genetic caste determination as well as future research opportunities are discussed.   Abstract Both gene flow and historical events influence the genetic diversity of natural populations. One way to understand their respective impact is to analyze population genetic structure at large spatial scales. We studied the distribution of genetic diversity of 17 populations of Vouacapoua americana (Caesalpiniaceae) in French Guiana, using nine micro satellite loci. Low genetic diversity was observed within populations, with a mean allelic richness and gene diversity of 4.1 and 0.506, respectively, which could be due to low effective population size and/or past bottlenecks. Using the regression between F-st/(1-F-st), estimated between pairs of populations, and the logarithm of the geographical distance, the spatial genetic structure can partly be explained by isolation-by-distance and limited gene flow among populations. This result is in agreement with the species' biology, including seed and pollen dispersal by rodents and insects, respectively. In contrast, no clear genetic signal of historical events was found when examining genetic differentiation among populations in relation to biogeographical hypotheses or by testing for bottlenecks within populations. Our conclusion is that nuclear spatial genetic structure of V. americana, at the geographic scale of French Guiana, is better explained by gene flow rather than by historical events.

  Abstract In this paper, we report a study of the mating system and gene flow of Symphonia globulifera, a hermaphroditic, mainly bird-pollinated tree species with a large geographic distribution in the tropical Americas and Africa. Using three microsatellites, we analysed 534 seeds of 28 open pollinated families and 164 adults at the experimental site 'Paracou' in French Guiana. We observed, compared to other tropical tree species, relatively high values for the effective number of alleles. Significant spatial genetic structure was detected, with trees at distances up to 150 m more genetically similar than expected at random. We estimated parameters of the mating system and gene flow by using the mixed mating model and the TwoGener approach. The estimated multilocus outcrossing rate, t(m), was 0.920. A significant level of biparental inbreeding and a high proportion of full-sibs were estimated for the 28 seed arrays. We estimated mean pollen dispersal distances between 27 and 53 m according to the dispersal models used. Although the adult population density of S. globulifera in Paracou was relatively high, the joint estimation of pollen dispersal and density of reproductive trees gave effective density estimates of 1.6 and 1.3 trees/ha.
