Abstract Despite their recognized essential role in soil, earthworms in tropical environments are still understudied. The aim of this study was to re-evaluate the diversity at the regional scale, as well as to investigate the environmental and spatial drivers of earthworm communities. We sampled earthworm communities across a range of habitats at six localities in French Guiana using three different sampling methods. We generated 1675 DNA barcodes and combined them with data from a previous study. Together, all sequences clustered into 119 MOTUs which were used as proxy to assess species richness. Only two MOTUs were common between the six localities and 20.2% were singletons, showing very high regional species richness and a high number of rare species. A canonical redundancy analysis was used to identify key drivers of the earthworm community composition. The RDA results and beta-diversity calculations both show strong species turnover and a strong spatial effect, resulting from dispersal limitations that are responsible for the current community composition. Sampling in different microhabitats allowed the discovery of 23 MOTUs that are exclusively found in decaying trunks and epiphytes, highlighting hidden diversity of earthworms outside of soil.

Abstract Differential growth response to light level is widely accepted as a potential mechanism for maintaining tree species richness in tropical forests. The position of tree species in the hierarchy of the canopy is considered an important indicator of species light capture and growth strategy. Paradoxically, the relative importance of species identity and competition for light in determining individual tree growth is poorly documented at the adult stage. In this study, we used a hierarchical Bayesian model to quantify the overall importance of species identity, light and belowground competition as determinants of tree growth in French Guiana tropical forest. Light competitive status is assessed by a crown exposure score and below ground competition is estimated from local crowding. We examined species sensitivity to both types of competition in relation to adult stature. Our results are based on annual diameter increments of more than 13,510 stems from 282 species monitored over 10years. Mean annual growth rate was 0.11cmy-1 with species identity explaining 35% of the individual variation in growth rate. Crown exposure and local crowding explained 3.5% and 2.4% of the variation in growth rate, respectively. Predicted changes in growth rate as crown exposure (resp. local crowding) index changed from lower to upper interquartile level was 0.03cmy-1 (resp. 0.02cmy-1). Species sensitivity to crown exposure and to local crowding were positively correlated (i) with predicted growth rate at high-light standardized conditions and (ii) with adult stature.This vertical niche partitioning is invoked to explain the limited contribution made by level of light competition for predicting individual tropical tree growth as the community-level response is dominated by the abundance of small-statured species with low sensitivity to light level.Light appears to drive the stem growth rate of tropical trees through species differentiation more than through individual tree growth limitation. This vertical stratification complements the previously reported regeneration niche and together these provide evidence for light niche partitioning in the three-dimensional space of tropical forests. © 2014 Elsevier B.V.

Abstract Analyses of tree diversity and community composition in tropical rain forests are usually based either on general herbarium data or on a restricted number of botanical plots. Despite their high taxonomic accuracy, both types of data are difficult to extrapolate to landscape scales. Meanwhile, forestry surveys provide quantitative occurrence data on large areas, and are thus increasingly used for landscape-scale analyses of tree diversity. However, the reliability of these approaches has been challenged because of the ambiguity of the common (vernacular) names used by foresters and the complexity of tree taxonomy in those hyper-diverse communities.We developed and tested a novel approach to evaluate taxonomic reliability of forestry surveys and to propagate the resulting uncertainty in the estimates of several diversity indicators (alpha and beta entropy, Fisher-alpha and Sørensen similarity). Our approach is based on Monte-Carlo processes that simulate communities by taking into account the expected accuracy and reliability of common names. We tested this method in French Guiana, on 9 one-hectare plots (4279 trees – DBH. ≥. 10. cm) for which both common names and standardized taxonomic determinations were available. We then applied our method of community simulation on large forestry inventories (560. ha) at the landscape scale and compared the diversity indices obtained for 10 sites with those computed from precise botanical determination situated at the same localities.We found that taxonomic reliability of forestry inventories varied from 22% (species level) to 83% (family level) in this Amazonian region. Indices computed directly with raw forestry data resulted in incorrect values, except for Gini-Simpson beta-diversity. On the contrary, our correction method provides more accurate diversity estimates, highly correlated with botanical measurements, for almost all diversity indices at both regional and local scales. We obtained a robust ranking of sites consistent with those shown by botanical inventories.These results show that (i) forestry inventories represent a significant part of taxonomic information, (ii) the relative diversity of regional sites can be successfully ranked using forestry inventory data using our method and (iii) forestry inventories can valuably contribute to the detection of large-scale diversity patterns when biases are well-controlled and corrected.The tools we developed as R-functions are available in supplementary material and can be adapted with local parameters to be used for forest management and conservation issues in other regional contexts. © 2014 Elsevier B.V.

Abstract The rise of integrative taxonomy, a multi-criteria approach used in characterizing species, fosters the development of new tools facilitating species delimitation. Mass spectrometric (MS) analysis of venom peptides from venomous animals has previously been demonstrated to be a valid method for identifying species. Here we aimed to develop a rapid chemotaxonomic tool for identifying ants based on venom peptide mass fingerprinting. The study focused on the biodiversity of ponerine ants (Hymenoptera: Formicidae: Ponerinae) in French Guiana. Initial experiments optimized the use of automated matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) to determine variations in the mass profiles of ant venoms using several MALDI matrices and additives. Data were then analyzed via a hierarchical cluster analysis to classify the venoms of 17 ant species. In addition, phylogenetic relationships were assessed and were highly correlated with methods using DNA sequencing of the mitochondrial gene cytochrome c oxidase subunit 1. By combining a molecular genetics approach with this chemotaxonomic approach, we were able to improve the accuracy of the taxonomic findings to reveal cryptic ant species within species complexes. This chemotaxonomic tool can therefore contribute to more rapid species identification and more accurate taxonomies. Biological significance: This is the first extensive study concerning the peptide analysis of the venom of both Pachycondyla and Odontomachus ants. We studied the venoms of 17 ant species from French Guiana that permitted us to fine-tune the venom analysis of ponerine ants via MALDI-TOF mass spectrometry. We explored the peptidomes of crude ant venom and demonstrated that venom peptides can be used in the identification of ant species. In addition, the application of this novel chemotaxonomic method combined with a parallel genetic approach using COI sequencing permitted us to reveal the presence of cryptic ants within both the Pachycondyla apicalis and Pachycondyla stigma species complexes. This adds a new dimension to the search for means of exploiting the enormous biodiversity of venomous ants as a source for novel therapeutic drugs or biopesticides. Abstract Tropical forests are the most diverse and productive ecosystems on Earth. While better understanding of these forests is critical for our collective future, until quite recently efforts to measure and monitor them have been largely disconnected. Networking is essential to discover the answers to questions that transcend borders and the horizons of funding agencies. Here we show how a global community is responding to the challenges of tropical ecosystem research with diverse teams measuring forests tree-by-tree in thousands of long-term plots. We review the major scientific discoveries of this work and show how this process is changing tropical forest science. Our core approach involves linking long-term grassroots initiatives with standardized protocols and data management to generate robust scaled-up results. By connecting tropical researchers and elevating their status, our Social Research Network model recognises the key role of the data originator in scientific discovery. Conceived in 1999 with RAINFOR (South America), our permanent plot networks have been adapted to Africa (AfriTRON) and Southeast Asia (T-FORCES) and widely emulated worldwide. Now these multiple initiatives are integrated via ForestPlots.net cyber-infrastructure, linking colleagues from 54 countries across 24 plot networks. Collectively these are transforming understanding of tropical forests and their biospheric role. Together we have discovered how, where and why forest carbon and biodiversity are responding to climate change, and how they feedback on it. This long-term pan-tropical collaboration has revealed a large long-term carbon sink and its trends, as well as making clear which drivers are most important, which forest processes are affected, where they are changing, what the lags are, and the likely future responses of tropical forests as the climate continues to change. By leveraging a remarkably old technology, plot networks are sparking a very modern revolution in tropical forest science. In the future, humanity can benefit greatly by nurturing the grassroots communities now collectively capable of generating unique, long-term understanding of Earth's most precious forests.

Abstract Production, emission, and absorption of biogenic volatile organic compounds (BVOCs) in ecosystem soils and associated impacts of nutrient availability are unclear; thus, predictions of effects of global change on source-sink dynamic under increased atmospheric N deposition and nutrition imbalances are limited. Here, we report the dynamics of soil BVOCs under field conditions from two undisturbed tropical rainforests from French Guiana. We analyzed effects of experimental soil applications of nitrogen (N), phosphorus (P), and N + P on soil BVOC exchanges (in particular of total terpenes, monoterpenes, and sesquiterpenes), to determine source and sink dynamics between seasons (dry and wet) and elevations (upper and lower elevations corresponding to top of the hills (30 m high) and bottom of the valley). We identified 45 soil terpenoids compounds emitted to the atmosphere, comprising 26 monoterpenes and 19 sesquiterpenes; of these, it was possible to identify 13 and 7 compounds, respectively. Under ambient conditions, soils acted as sinks of these BVOCs, with greatest soil uptake recorded for sesquiterpenes at upper elevations during the wet season (-282 μg m-2 h-1). Fertilization shifted soils from a sink to source, with greatest levels of terpene emissions recorded at upper elevations during the wet season, following the addition of N (monoterpenes: 406 μg m-2 h-1) and P (sesquiterpenes: 210 μg m-2 h-1). Total soil terpene emission rates were negatively correlated with total atmospheric terpene concentrations. These results indicate likely shifts in tropical soils from sink to source of atmospheric terpenes under projected increases in N deposition under global change, with potential impacts on regional-scale atmospheric chemistry balance and ecosystem function.

Abstract Biological nitrogen fixation (BNF) is a fundamental part of nitrogen cycling in tropical forests, yet little is known about the contribution made by free-living nitrogen fixers inhabiting the often-extensive forest canopy. We used the acetylene reduction assay, calibrated with 15N2, to measure free-living BNF on forest canopy leaves, vascular epiphytes, bryophytes and canopy soil, as well as on the forest floor in leaf litter and soil. We used a combination of calculated and published component densities to upscale free-living BNF rates to the forest level. We found that bryophytes and leaves situated in the canopy in particular displayed high mass-based rates of free-living BNF. Additionally, we calculated that nearly 2 kg of nitrogen enters the forest ecosystem through free-living BNF every year, 40% of which was fixed by the various canopy components. Our results reveal that in the studied tropical lowland forest a large part of the nitrogen input through free-living BNF stems from the canopy, but also that the total nitrogen inputs by free-living BNF are lower than previously thought and comparable to the inputs of reactive nitrogen by atmospheric deposition.

Abstract Selective logging in tropical forests is often perceived as a source of forest degradation and carbon emissions. Improved practices, such as reduced-impact logging (RIL), and alternative timber production strategies (e.g. plantations) can drastically change the overall carbon impact of the wood production sector. Assessing the carbon balance of timber production is crucial but highly dependent on methodological approaches, especially regarding system boundaries and temporality. We developed a temporally-explicit and territory scale model of carbon balance calibrated with long-term local data using Bayesian inference. The model accounts for carbon fluxes from selective logging in natural forest, timber plantation, first transformation and avoided emissions through energy substitution. We used it to compare prospective scenarios of development for the wood sector in French Guiana. Results show that intensification of practices, through increased logging intensity conducted with RIL and establishment of timber plantations, are promising development strategies to reduce the carbon emissions of the French-Guianese wood sector, as well as the area needed for wood production and hence the pressure on natural forests. By reducing logging damage by nearly 50%, RIL allows increasing logging intensity in natural forest from 20 m3 ha−1 to 30 m3 ha−1 without affecting the carbon balance. The use of logging byproducts as fuelwood also improved the carbon balance of selective logging, when substituted to fossil fuel. Allocating less than 30 000 ha to plantation would allow producing 200 000 m3 of timber annually, while the same production in natural forest would imply logging more than 400 000 ha over 60 years. Timber plantation should be preferentially established on non-forested lands, as converting natural forests to plantation leads to high carbon emission peak over the first three decades. We recommend a mixed-strategy combining selective logging in natural forests and plantations as a way to improve long-term carbon balance while reducing short-term emissions. This strategy can reduce the pressure on natural forests while mitigating the risks of changing practices and allowing a diversified source of timber for a diversity of uses. It requires adaptation of the wood sector and development of technical guidelines. Research and monitoring efforts are also needed to assess the impacts of changing practices on other ecosystem services, especially biodiversity conservation.

Abstract Cytogenetic techniques provide powerful insights on species-rich taxa–such as ants–allowing better understanding of their biodiversity. Some hints on evolutionary paths can be observed through comparative populational cytogenetics among different ant groups. In this study, the karyotype of Strumigenys louisianae Roger from the Amazon rainforest is described and showed diploid chromosome number of 26 chromosomes. This configuration intriguingly contrasts with the already described karyotype for this species from the Atlantic rainforest with only 2n = 4 chromosomes. 18S rDNA site were detected on the pericentromeric region of the long arm of a metacentric pair and co-localizing with GC-rich chromatin. Recurrent synonymizations of S. louisianae may not reflect the species status of this taxon. The karyotypic differences and the observable morphological variation between the populations of both localities corroborates the idea of a species complex within S. louisianae. The morphology of S. louisianae from the Amazonian region is similar to that from the United States, the type locality. On the other hand, specimens from the Atlantic rainforest are more similar to the junior synonym Strumigenys unidentata Mayr. This study reinforces the need of taxonomical revision in S. louisianae by means of integrative taxonomy approaches.

Abstract In the face of increasing drug resistance, the development of new anthelmintics is critical for controlling nematodes that parasitise livestock. Although hymenopteran venom toxins have attracted attention for applications in agriculture and medicine, few studies have explored their potential as anthelmintics. Here we assessed hymenopteran venoms as a possible source of new anthelmintic compounds by screening a panel of ten hymenopteran venoms against Haemonchus contortus, a major pathogenic nematode of ruminants. Using bioassay-guided fractionation coupled with liquid chromatography-tandem mass spectrometry, we identified four novel anthelmintic peptides (ponericins) from the venom of the neotropical ant Neoponera commutata and the previously described ponericin M−PONTX−Na1b from Neoponera apicalis venom. These peptides inhibit H. contortus development with IC50 values of 2.8–5.6 μM. Circular dichroism spectropolarimetry indicated that the ponericins are unstructured in aqueous solution but adopt α-helical conformations in lipid mimetic environments. We show that the ponericins induce non-specific membrane perturbation, which confers broad-spectrum antimicrobial, insecticidal, cytotoxic, hemolytic, and algogenic activities, with activity across all assays typically correlated. We also show for the first time that ponericins induce spontaneous pain behaviour when injected in mice. We propose that the broad-spectrum activity of the ponericins enables them to play both a predatory and defensive role in neoponeran ants, consistent with their high abundance in venom. This study reveals a broader functionality for ponericins than previously assumed, and highlights both the opportunities and challenges in pursuing ant venom peptides as potential therapeutics.
