Abstract Key message: Genetic diversity appears to be unaffected by disturbance in a stand of the light-demanding Neotropical treeV. michelii. Although spatial genetic structure is modified in post-disturbance cohorts, mixing of seeds from different mother trees in canopy gaps appears to efficiently maintain genetic admixture. Context: The interplay between genetic and demographic processes has major consequences on population viability. Population size affects demographic trends, while genetic diversity insures viability by reducing risks of inbreeding depression and by maintaining adaptive potential. Yet, the consequences of increases in census size (as opposed to effective size) on genetic diversity of forest populations are poorly known. Aims: We have studied the structure of genetic diversity in populations of saplings of the light-responsive tree, Virola michelii (Myristicaceae, the nutmeg family), in two plots having undergone different levels of canopy-gap opening disturbance. This allowed us to test the “intermediate disturbance” hypothesis, which generally applies to species diversity, at the intra-specific scale. Methods: Levels and distribution of genetic diversity were compared between plots and between life stages. Sapling parentage was analysed to infer each adult tree’s contribution to regeneration. Results: Genetic diversity was higher, and spatial genetic structure was stronger in the post-disturbance than in the control seedling population. Parentage analysis suggested that a limited number of parents contributed to most of the regeneration, but that efficient mixing of their progeny may have enhanced the diversity of saplings occupying canopy gaps. Conclusion: A mixture of demo-genetic processes may contribute to maintain genetic diversity in spite of, or possibly due to, ecosystem disturbance in V. michelii. © 2015, INRA and Springer-Verlag France.

Abstract Patterns of leaf production and leaf fall directly influence leaf area index and forest productivity. Here, we focused on Cecropia sciadophylla individuals inhabiting the extremes of the gradient in seasonality in rainfall at which C. sciadophylla occurs. In Colombia and French Guiana we compared the intra-annual variation in leaf production as well as the intra-annual fluctuation in internode length on a total of 69 saplings ranging in size from 1 to 2 m. The mean rate of leaf production was ~2 leaves mo -1 in both populations, and the rate of leaf production was constant throughout the year. Our results showed monthly variation in internode length and the number of live leaves per sapling in the seasonal habitat and variation only in internode length in the everwet habitat. Because the rate of leaf production is constant at both localities, the difference in number of live leaves per sapling at the seasonal site must reflect seasonal variation in leaf life span. We show that in Cecropia, internode length can serve as an indicator of precipitation seasonality. Finally an open question is whether leaf production in other pioneer species is also independent of climatic seasonal cues. This information could allow us to link growth and climate of secondary forest species and better understand how past and future climate can affect plant growth trajectories. © Cambridge University Press 2013.

Abstract Economic globalization manifests in landscapes through regional integration initiatives involving trans-boundary infrastructure. While the relationships of roads, accessibility and land cover are well-understood, they have rarely been considered across borders in national frontier regions. We therefore pursue an analysis of infrastructure connectivity and land cover change in the tri-national frontier of the southwestern Amazon where Bolivia, Brazil and Peru meet, and where the Inter-Oceanic Highway has recently been paved. We integrate satellite, survey, climate and other data for a sample of rural communities that differ in terms of highway paving across the tri-national frontier. We employ a suite of explanatory variables tied to road paving and other factors that vary both across and within the three sides of the frontier in order to model their importance for deforestation. A multivariate analysis of non-forest land cover during 2005-2010 confirms the importance of paving status and travel times, as well as land tenure and other factors. These findings indicate that integration affects land cover, but does not eliminate the effects of other factors that vary across the frontier, which bears implications for the study of globalization, trans-boundary infrastructure, environmental governance and land cover change. © 2013 Elsevier Ltd.

Abstract In the mutualisms involving the myrmecophyte Cecropia obtusa and Azteca ovaticeps or A. alfari, both predatory, the ants defend their host trees from enemies and provide them with nutrients (myrmecotrophy). A. ovaticeps provisioned with prey and then 15N-enriched food produced more individuals than did control colonies (not artificially provisioned). This was not true for A. alfari colonies, possibly due to differences in the degree of maturity of the colonies for the chosen range of host tree sizes (less than 3 m in height). Myrmecotrophy was demonstrated for both Azteca species as provisioning the ants with 15N-enriched food translated into higher δ 15N values in host plant tissues, indicating that nitrogen passed from the food to the plant. Thus, the predatory activity of their guest ants benefits the Cecropia trees not only because the ants protect them from defoliators since most prey are phytophagous insects but also because the plant absorbs nutrients. © 2012 Académie des sciences. Abstract Guadua sarcocarpa and Guadua weberbaueri (Poaceae: Bambuseae) have a negative influence on tree regeneration and recruitment in bamboo-dominated forests of southwestern Amazonia. The lack of advanced regeneration and sparse canopy in this forest type present a considerable challenge for developing sustainable timber management plans. We conducted field studies in the Porto Dias Agroextractive Settlement Project in Acre, Brazil to assess influences of logging in bamboo-dominated forest sites. Taxonomic composition, stand structure, aboveground biomass, commercial timber volume, and commercial tree seedling and bamboo culm density were compared between five logged vs. unlogged sites in different landholdings, using modified 0.5. ha Gentry plots. No differences in taxonomic composition, aboveground biomass, adult and juvenile stem density, or woody seedling and bamboo culm density were detected between paired logged and unlogged sites. Commercial timber volume, however, was reduced by almost two-thirds in logged plots, suggesting that long-term timber management goals in this forest type are compromised since so few future crop trees remained onsite. Our findings indicate that in order to maximize local management objectives, community forest managers must approach logging in bamboo-dominated forests with caution. We suggest an integration of non-timber forest product extraction with low harvest intensity and low-impact logging, tending of natural regeneration, and diversification of commercial species. © 2014 Elsevier B.V.

Title Identification and characterization of a novel antimicrobial peptide from the venom of the ant Tetramorium bicarinatum Type Journal Article

Abstract A novel antimicrobial peptide, named Bicarinalin, has been isolated from the venom of the ant Tetramorium bicarinatum. Its amino acid sequence has been determined by de novo sequencing using mass spectrometry and by Edman degradation. Bicarinalin contained 20 amino acid residues and was C-terminally amidated as the majority of antimicrobial peptides isolated to date from insect venoms. Interestingly, this peptide had a linear structure and exhibited no meaningful similarity with any known peptides. Antibacterial activities against Staphylococcus aureus and S. xylosus strains were evaluated using a synthetic replicate. Bicarinalin had a potent and broad antibacterial activity of the same magnitude as Melittin and other hymenopteran antimicrobial peptides such as Pilosulin or Defensin. Moreover, this antimicrobial peptide has a weak hemolytic activity compared to Melittin on erythrocytes, suggesting potential for development into an anti-infective agent for use against emerging antibiotic-resistant pathogens. © 2012 Elsevier Inc.

Abstract Investigations were carried out on the efficacy of extracts from seven Amazonian woods (Bagassa guianensis, Manilkara huberi, Sextonia rubra, Vouacapoua americana, Andira surinamensis, Handroanthus serratifolius, and Qualea rosea) with varying natural durability to reduce soft-rot degradation in a 6-wk soil-bed test. Six of the wood extracts had shown efficacy against soft-rot fungi. In particular, the preservation efficacies of B. guianensis, H. serratifolius, and S. rubra extracts were highly significant up to retention levels of 23, 25, and 12 kg m-3, respectively. Three extracts (A. surinamensis, H. serratifolius, and Q. rosea) were then tested against Gloeophyllum trabeum (brown rot) and Trametes versicolor (white rot), in an agar-block test. H. serratifolius wood extract was very efficient at protecting P. sylvestris samples at 5.1 kg m-3 against the brown rot. This extract could be used as a basis for new wood protectant formulations. © 2012 Elsevier Ltd.

Abstract Eperua falcata (Aublet), a late-successional species in tropical rainforest and one of the most abundant tree in French Guiana, has developed an original strategy concerning N-acquisition by largely preferring nitrate, rather than ammonium (H. Schimann, S. Ponton, S. Hättenschwiler, B. Ferry, R. Lensi, A.M. Domenach, J.C. Roggy, Differing nitrogen use strategies of two tropical rainforest tree species in French Guiana: evidence from 15N natural abundance and microbial activities, Soil Biol. Biochem. 40 (2008) 487-494). Given the preference of this species for nitrate, we hypothesized that root exudates would promote nitrate availability by (a) enhancing nitrate production by stimulating ammonium oxidation or (b) minimizing nitrate losses by inhibiting denitrification.Root exudates were collected in situ in monospecific planted plots. The phytochemical analysis of these exudates and of several of their corresponding root extracts was achieved using UHPLC/DAD/ESI-QTOF and allowed the identification of diverse secondary metabolites belonging to the flavonoid family.Our results show that (i) the distinct exudation patterns observed are related to distinct root morphologies, and this was associated with a shift in the root flavonoid content, (ii) a root extract representative of the diverse compounds detected in roots showed a significant and selective metabolic inhibition of isolated denitrifiers invitro, and (iii) in soil plots the abundance of nirK-type denitrifiers was negatively affected in rhizosphere soil compared to bulk. Altogether this led us to formulate hypothesis concerning the ecological role of the identified compounds in relation to N-acquisition strategy of this species. © 2013 Elsevier Masson SAS.

Abstract High-speed power-amplification mechanisms are common throughout the animal kingdom. In ants, power-amplified trap-jaw mandibles have evolved independently at least four times, including once in the subfamily Ponerinae which contains the sister genera Odontomachus and Anochetus. In Odontomachus, mandible strikes have been relatively well described and can occur in <0.15 ms and reach speeds of over 60 m s−1. In contrast, the kinematics of mandible strikes have not been examined in Anochetus, whose species are smaller and morphologically distinct from Odontomachus. In this study, we describe the mandible strike kinematics of four species of Anochetus representative of the morphological, phylogenetic, and size diversity present within the genus. We also compare their strikes to two representative species of Odontomachus. We found that two species, Anochetus targionii and Anochetus paripungens, have mandible strikes that overall closely resemble those found in Odontomachus, reaching a mean maximum rotational velocity and acceleration of around 3.7 × 104 rad s−1 and 8.5 × 108 rad s−2, respectively. This performance is consistent with predictions based on body size scaling relationships described for Odontomachus. In contrast, Anochetus horridus and Anochetus emarginatus have slower strikes relative to the other species of Anochetus and Odontomachus, reaching mean maximum rotational velocity and acceleration of around 1.3 × 104 rad s−1 and 2 × 108 rad s−2, respectively. This variation in strike performance among species of Anochetus likely reflects differences in evolutionary history, physiology, and natural history among species. © 2018 The Zoological Society of London

Abstract Background Most ant venoms consist predominantly of small linear peptides, although some contain disulfide-linked peptides as minor components. However, in striking contrast to other ant species, some Anochetus venoms are composed primarily of disulfide-rich peptides. In this study, we investigated the venom of the ant Anochetus emarginatus with the aim of exploring these novel disulfide-rich peptides. Methods The venom peptidome was initially investigated using a combination of reversed-phase HPLC and mass spectrometry, then the amino acid sequences of the major peptides were determined using a combination of Edman degradation and de novo MS/MS sequencing. We focused on one of these peptides, U1-PONTX-Ae1a (Ae1a), because of its novel sequence, which we predicted would form a novel 3D fold. Ae1a was chemically synthesized using Fmoc chemistry and its 3D structure was elucidated using NMR spectroscopy. The peptide was then tested for insecticidal activity and its effect on a range of human ion channels. Results Seven peptides named poneritoxins (PONTXs) were isolated and sequenced. The three-dimensional structure of synthetic Ae1a revealed a novel, compact scaffold in which a C-terminal β-hairpin is connected to the N-terminal region via two disulfide bonds. Synthetic Ae1a reversibly paralyzed blowflies and inhibited human L-type voltage-gated calcium channels (CaV1). Conclusions Poneritoxins from Anochetus emarginatus venom are a novel class of toxins that are structurally unique among animal venoms. General significance This study demonstrates that Anochetus ant venoms are a rich source of novel ion channel modulating peptides, some of which might be useful leads for the development of biopesticides. © 2016
