Abstract: A critical hurdle in ant venom proteomic investigations is the lack of databases to comprehensively and specifically identify the sequence and function of venom proteins and peptides. To resolve this, we used venom gland transcriptomics to generate a sequence database that was used to assign the tandem mass spectrometry (MS) fragmentation spectra of venom peptides and proteins to specific transcripts. This was performed alongside a shotgun liquid chromatography-mass spectrometry (LC-MS/MS) analysis of the venom to confirm that these assigned transcripts were expressed as proteins. Through the combined transcriptomic and proteomic investigation of Paraponera clavata venom, we identified four times the number of proteins previously identified using 2D-PAGE alone. In addition to this, by mining the transcriptomic data, we identified several novel peptide sequences for future pharmacological investigations, some of which conform with inhibitor cysteine knot motifs. These types of peptides have the potential to be developed into pharmaceutical or bioinsecticide peptides. © 2020 by the authors. Licensee MDPI, Basel, Switzerland.

Abstract: Recent studies have shown that the inner bark is implicated in the postural control of inclined tree stems through the interaction between wood radial growth and tangential expansion of a trellis fiber network in bark. Assessing the taxonomic extent of this mechanism requires a screening of the diversity in bark anatomy and mechanical stress. The mechanical state of bark was measured in 15 tropical tree species from various botanical families on vertical mature trees, and related to the anatomical structure of the bark. Significant tensile or compressive longitudinal stresses were observed in the stems of most species. Tensile longitudinal stress was observed in various botanical families and was always associated with fibers arranged in a trellis-like structure and strong dilatation of rays. The highest tensile stress was recorded in species with gelatinous fibers forming a treillis. Compressive stress was typically associated with a large amount of sclereids in the bark, supporting the differentiation of sclereids as a potential origin of the generation of longitudinal compressive stresses in bark. In species exhibiting both a fibrous trellis structure and a significant amount of sclereids, the sign of longitudinal stress may depend on the balance between these two mechanisms.

Abstract: The FLUXNET2015 dataset provides ecosystem-scale data on CO2, water, and energy exchange between the biosphere and the atmosphere, and other meteorological and biological measurements, from 212 sites around the globe (over 1500 site-years, up to and including year 2014). These sites, independently managed and operated, voluntarily contributed their data to create global datasets. Data were quality controlled and processed using uniform methods, to improve consistency and intercomparability across sites. The dataset is already being used in a number of applications, including ecophysiology studies, remote sensing studies, and development of ecosystem and Earth system models. FLUXNET2015 includes derived-data products, such as gap-filled time series, ecosystem respiration and photosynthetic uptake estimates, estimation of uncertainties, and metadata about the measurements, presented for the first time in this paper. In addition, 206 of these sites are for the first time distributed under a Creative Commons (CC-BY 4.0) license. This paper details this enhanced dataset and the processing methods, now made available as open-source codes, making the dataset more accessible, transparent, and reproducible.

Abstract: The intensification of dry seasons is a major threat to freshwater biodiversity in Neotropical regions. Little is known about resistance to drying stress and the underpinning traits in Neotropical freshwater species, so we don't know whether desiccation resistance allows to anticipate shifts in biological diversity under future climate scenarios. Here, we used the aquatic invertebrates that live in the rainwater-filled leaves of tank bromeliads, to examine the extent to which desiccation resistance of species measured in the laboratory predicts community response to drought intensification in nature. We measured desiccation resistance in 17 invertebrate species (>90% of the biomass usually found in bromeliads of French Guiana) by recording the median lethal time (LT50) of experimental populations exposed to controlled conditions of residual moisture. In the field, we placed rainshelters above tank bromeliads to emulate drought scenarios ranging from the ambient norm to IPCC scenarios and extreme events, and we recorded the response of functional community structure. LT50 ranged from 4.18 to 19.06 days, and was related to cuticle content and dry body mass. Among other functional indicators that represent strategies to optimize resource use under stressful conditions (e.g., habitat use, trophic specialization), LT50 was the best predictor of community structure responses along a gradient of emulated drought intensities. Therefore, species’ LT50s measured under laboratory conditions can be used to forecast aquatic community response to drying stress in nature. Anticipating how species will cope with drought has never been more important for environmental managers to support climate change adaptation. We show that desiccation resistance in freshwater invertebrates is a key indicator of potential population size and local–global range shifts, and this could be especially true in the Neotropics where species have narrow physiological tolerances for climatic variation. © 2020 Elsevier Ltd

Abstract: Background and aims: Biological fixation of atmospheric nitrogen (N2) is the main pathway for introducing N into unmanaged ecosystems. While recent estimates suggest that free-living N fixation (FLNF) accounts for the majority of N fixed in mature tropical forests, the controls governing this process are not completely understood. The aim of this study was to quantify FLNF rates and determine its drivers in two tropical pristine forests of French Guiana. Methods: We used the acetylene reduction assay to measure FLNF rates at two sites, in two seasons and along three topographical positions, and used regression analyses to identify which edaphic explanatory variables, including carbon (C), nitrogen (N), phosphorus (P) and molybdenum (Mo) content, pH, water and available N and P, explained most of the variation in FLNF rates. Results: Overall, FLNF rates were lower than measured in tropical systems elsewhere. In soils seasonal variability was small and FLNF rates differed among topographies at only one site. Water, P and pH explained 24% of the variation. In leaf litter, FLNF rates differed seasonally, without site or topographical differences. Water, C, N and P explained 46% of the observed variation. We found no regulatory role of Mo at our sites. Conclusions: Rates of FLNF were low in primary rainforest on poor soils on the Guiana shield. Water was the most important rate-regulating factor and FLNF increased with increasing P, but decreased with increasing N. Our results support the general assumption that N fixation in tropical lowland forests is limited by P availability. © 2019, The Author(s).

Abstract: Differences in photosynthetic capacity among tree species and tree functional types are currently assumed to be largely driven by variation in leaf nutrient content, particularly nitrogen (N). However, recent studies indicate that leaf N content is often a poor predictor of variation in photosynthetic capacity in tropical trees. In this study, we explored the relative importance of area-based total leaf N content (Ntot) and within-leaf N allocation to photosynthetic capacity versus light-harvesting in controlling the variation in photosynthetic capacity (i.e. Vcmax, Jmax) among mature trees of 12 species belonging to either early (ES) or late successional (LS) groups growing in a tropical montane rainforest in Rwanda, Central Africa. Photosynthetic capacity at a common leaf temperature of 25˚C (i.e. maximum rates of Rubisco carboxylation, Vcmax25 and of electron transport, Jmax25) was higher in ES than in LS species (+ 58% and 68% for Vcmax25 and Jmax25, respectively). While Ntot did not significantly differ between successional groups, the photosynthetic dependency on Ntot was markedly different. In ES species, Vcmax25 was strongly and positively related to Ntot but this was not the case in LS species. However, there was no significant trade-off between relative leaf N investments in compounds maximizing photosynthetic capacity versus compounds maximizing light harvesting. Both leaf dark respiration at 25˚C (+ 33%) and, more surprisingly, apparent photosynthetic quantum yield (+ 35%) was higher in ES than in LS species. Moreover, Rd25 was positively related to Ntot for both ES and LS species. Our results imply that efforts to quantify carbon fluxes of tropical montane rainforests would be improved if they considered contrasting within-leaf N allocation and photosynthetic Ntot dependencies between species with different successional strategies. © Copyright © 2020 Ziegler, Dusenge, Nyirambangutse, Zibera, Wallin and Uddling.

Abstract: Soil water and nutrient availability are key drivers of tree species distribution and forest ecosystem functioning, with strong species differences in water and nutrient use. Despite growing evidence for intraspecific trait differences, it remains unclear under which circumstances the effects of environmental gradients trump those of ontogeny and taxonomy on important functional dimensions related to resource use, particularly in tropical forests. Here, we explore how physiological, chemical, and morphological traits related to resource use vary between life stages in four species within the genus Micropholis that is widespread in lowland Amazonia. Specifically, we evaluate how environment, developmental stage, and taxonomy contribute to single-trait variation and multidimensional functional strategies. We find that environment, developmental stage, and taxonomy differentially contribute to functional dimensions. Habitats and seasons shape physiological and chemical traits related to water and nutrient use, whereas developmental stage and taxonomic identity impact morphological traits –especially those related to the leaf economics spectrum. Our findings suggest that combining environment, ontogeny, and taxonomy allows for a better understanding of important functional dimensions in tropical trees and highlights the need for integrating tree physiological and chemical traits with classically used morphological traits to improve predictions of tropical forests’ responses to environmental change. © 2019 The Authors New Phytologist © 2019 New Phytologist Trust

Abstract: The rainforests of the Neotropics shelter a vast diversity of plant, animal and microscopic species that provide critical ecosystem goods and services for both local and worldwide populations. These environments face a major crisis due to increased deforestation, pollution, and climate change, emphasizing the need for more effective conservation efforts. The adequate monitoring of these ecosystems has proven a complex and time consuming endeavour, which depends on ever dwindling taxonomic expertise. To date, many species remain undiscovered, let alone described, with otherwise limited information regarding known species population distributions and densities. Overcoming these knowledge shortfalls and practical limitations is becoming increasingly possible through techniques based on environmental DNA (eDNA), i.e., DNA that can be obtained from environmental samples (e.g. tissues, soil, sediment, water, etc.). When coupled with high-throughput sequencing, these techniques now enable realistic, cost-effective, and standardisable biodiversity assessments. This opens up enormous opportunities for advancing our understanding of complex and species-rich tropical communities, but also in facilitating large-scale biomonitoring programs in the neotropics. In this review, we provide a brief introduction to eDNA methods, and an overview of their current and potential uses in both terrestrial and aquatic ecosystems of neotropical rainforests. We also discuss the limits and challenges of these methods for our understanding and monitoring of biodiversity, as well as future research and applied perspectives of these techniques in neotropical rainforests, and beyond. © 2020 Elsevier Ltd

Abstract: There is growing recognition that ecosystems may be more impacted by infrequent extreme climatic events than by changes in mean climatic conditions. This has led to calls for experiments that explore the sensitivity of ecosystems over broad ranges of climatic parameter space. However, because such response surface experiments have so far been limited in geographic and biological scope, it is not clear if differences between studies reflect geographic location or the ecosystem component considered. In this study, we manipulated rainfall entering tank bromeliads in seven sites across the Neotropics, and characterized the response of the aquatic ecosystem in terms of invertebrate functional composition, biological stocks (total invertebrate biomass, bacterial density) and ecosystem fluxes (decomposition, carbon, nitrogen). Of these response types, invertebrate functional composition was the most sensitive, even though, in some sites, the species pool had a high proportion of drought-tolerant families. Total invertebrate biomass was universally insensitive to rainfall change because of statistical averaging of divergent responses between functional groups. The response of invertebrate functional composition to rain differed between geographical locations because (1) the effect of rainfall on bromeliad hydrology differed between sites, and invertebrates directly experience hydrology not rainfall and (2) the taxonomic composition of some functional groups differed between sites, and families differed in their response to bromeliad hydrology. These findings suggest that it will be difficult to establish thresholds of “safe ecosystem functioning” when ecosystem components differ in their sensitivity to climatic variables, and such thresholds may not be broadly applicable over geographic space. In particular, ecological forecast horizons for climate change may be spatially restricted in systems where habitat properties mediate climatic impacts, and those, like the tropics, with high spatial turnover in species composition. © 2020 by the Ecological Society of America

Abstract: The 2015–2016 El Niño event ranks as one of the most severe on record in terms of the magnitude and extent of sea surface temperature (SST) anomalies generated in the tropical Pacific Ocean. Corresponding global impacts on the climate were expected to rival, or even surpass, those of the 1997–1998 severe El Niño event, which had SST anomalies that were similar in size. However, the 2015–2016 event failed to meet expectations for hydrologic change in many areas, including those expected to receive well above normal precipitation. To better understand how climate anomalies during an El Niño event impact soil moisture, we investigate changes in soil moisture in the humid tropics (between ±25∘) during the three most recent super El Niño events of 1982–1983, 1997–1998 and 2015–2016, using data from the Global Land Data Assimilation System (GLDAS). First, we use in situ soil moisture observations obtained from 16 sites across five continents to validate and bias-correct estimates from GLDAS (r2=0.54). Next, we apply a k-means cluster analysis to the soil moisture estimates during the El Niño mature phase, resulting in four groups of clustered data. The strongest and most consistent decreases in soil moisture occur in the Amazon basin and maritime southeastern Asia, while the most consistent increases occur over eastern Africa. In addition, we compare changes in soil moisture to both precipitation and evapotranspiration, which showed a lack of agreement in the direction of change between these variables and soil moisture most prominently in the southern Amazon basin, the Sahel and mainland southeastern Asia. Our results can be used to improve estimates of spatiotemporal differences in El Niño impacts on soil moisture in tropical hydrology and ecosystem models at multiple scales.
