  Abstract In French Guiana, Quassia amara L. (Simaroubaceae) leaf tea is a well-known widely used traditional antimalarial remedy. Impact of the vegetal sampling condition on in vivo and in vitro antimalarial activity was assessed. Traditional infusions were prepared with juvenile or mature leaves, both either fresh or dried. Results showed that growing stage and freshness of vegetal material exert a striking effect on antimalarial activity, both in vitro and in vivo. By far, leaf tea made from fresh juvenile (FJ) Quassia amara leaves was the most active. In vitro, active component (simalikalactone D) concentration correlates biological activities, although unexplained subtle variations were observed. In vivo, tea made with dried juvenile (DJ) leaves displays a peculiar behavior, meaning that some components may help simalikalactone D delivery or may be active in vivo only, therefore enhancing the expected curative effect of the traditional preparation.   Abstract Quassia amara L. (Simaroubaceae) is a species widely used as tonic and is claimed to be an efficient antimalarial all over the Northern part of the Amazon basin. Quassinoid compound Simalikalactone D (SkD) has been shown to be one of the molecules responsible for the antiplasmodial activity of a watery preparation made out of juvenile fresh leaves of this plant. Because of its strong antimalarial activity, we decided to have a further insight of SkD pharmacological properties, alone or in association with classical antimalarials. At concentrations of up to 200 μM, we showed herein that SkD did not exert any apoptotic or necrotic activities in vitro on lymphoblastic cells. However, an antiproliferative effect was evident at concentrations higher than 45. nM. SkD was inefficient at inhibiting heme biomineralization and the new permeability pathways induced by the parasite in the host erythrocyte membrane. With respect to Plasmodium falciparum erythrocytic stages, SkD was almost inactive on earlier and later parasite stages, but potently active at the 30th h of parasite cycle when DNA replicates in mature trophozoites. In vitro combination studies with conventional antimalarial drugs showed that SkD synergizes with atovaquone (ATO). The activity of ATO on the Plasmodium mitochondrial membrane potential was enhanced by SkD, which on its own had a poor effect on this cellular parameter. © 2012 Elsevier Inc.

  Abstract French Guiana (North-East Amazonia) records high malaria incidence rates. The traditional antimalarial remedy most widespread there is a simple tea made out from Quassia amara L. leaves (Simaroubaceae). This herbal tea displays an excellent antimalarial activity both in vitro and in vivo. A known quassinoid, simalikalactone D (SkD), was identified as the active compound, with an IC50 value of 10 nM against FeB1 Plasmodium falciparum chloroquine resistant strain in vitro. Lastly, it inhibits 50% of Plasmodium yoelii yoelii rodent malaria parasite at 3.7 mg/kg/day in vivo by oral route. These findings confirm the traditional use of this herbal tea.   Abstract Ants figure prominently among the worst invasive species because of their enormous ecological and economic impacts. However, it remains to be investigated which species would be behaviourally dominant when confronted with another invasive ant species, should two species be introduced in the same area. In the future, many regions might have suitable environmental conditions for several invasive ant species, as predicted under climate change scenarios. Here, we explored interactions among several highly invasive ant species, which have been shown to have overlapping suitable areas. The aim of this study was to evaluate the performance in interference competition of seven of the world’s worst invasive ant species (Anoplolepis gracilipes, Paratrechina longicornis, Myrmica rubra, Linepithema humile, Lasius neglectus, Wasmannia auropunctata and Pheidole megacephala). We conducted pairwise confrontations, testing the behaviour of each species against each of the six other species (in total 21 dyadic confrontations). We used single worker confrontations and group interactions of 10 versus 10 individuals to establish a dominance hierarchy among these invasive ant species. We discovered two different behavioural strategies among these invasive ants: three species displayed evasive or indifferent behaviour when individuals or groups were confronted (A. gracilipes, Pa. longicornis, M. rubra), while the four remaining species were highly aggressive during encounters and formed a linear dominance hierarchy. These findings contrast with the widespread view that invasive ants form a homogeneous group of species displaying the ‘invasive syndrome’, which includes generally aggressive behaviour. The dominance hierarchy among the four aggressive species may be used to predict the outcome of future competitive interactions under some circumstances. Yet, the existence of several behavioural strategies renders such a prediction less straightforward.

  Abstract Phylogenetic patterns and the underlying speciation processes can be deduced from morphological, functional, and ecological patterns of species similarity and divergence. In some cases, though, species retain multiple similarities and remain almost indistinguishable; in other cases, evolutionary convergence can make such patterns misleading; very often in such cases, the “true” picture only emerges from carefully built molecular phylogenies, which may come with major surprises. In addition, closely related species may experience gene flow after divergence, thus potentially blurring species delimitation. By means of advanced inferential methods, we studied molecular divergence between species of the Virola genus (Myristicaceae): widespread Virola michelii and recently described, endemic V. kwatae, using widespread V. surinamensis as a more distantly related outgroup with different ecology and morphology—although with overlapping range. Contrary to expectations, we found that the latter, and not V. michelii, was sister to V. kwatae. Therefore, V. kwatae probably diverged from V. surinamensis through a recent morphological and ecological shift, which brought it close to distantly related V. michelii. Through the modeling of the divergence process, we inferred that gene flow between V. surinamensis and V. kwatae stopped soon after their divergence and resumed later, in a classical secondary contact event which did not erase their ecological and morphological differences. While we cannot exclude that initial divergence occurred in allopatry, current species distribution and the absence of geographical barriers make complete isolation during speciation unlikely. We tentatively conclude that (a) it is possible that divergence occurred in allopatry/parapatry and (b) secondary contact did not suppress divergence. © 2020 The Authors.   Abstract Bacteria living on the cuticle of ants are generally studied for their protective role against pathogens, especially in the clade of fungus-growing ants. However, little is known regarding the diversity of cuticular bacteria in other ant host species, as well as the mechanisms leading to the composition of these communities. Here, we used 16S rRNA gene amplicon sequencing to study the influence of host species, species interactions and the pool of bacteria from the environment on the assembly of cuticular bacterial communities on two phylogenetically distant Amazonian ant species that frequently nest together inside the roots system of epiphytic plants, Camponotus femoratus and Crematogaster levior. Our results show that (a) the vast majority of the bacterial community on the cuticle is shared with the nest, suggesting that most bacteria on the cuticle are acquired through environmental acquisition, (b) 5.2% and 2.0% of operational taxonomic units (OTUs) are respectively specific to Ca. femoratus and Cr. levior, probably representing their respective core cuticular bacterial community, and (c) 3.6% of OTUs are shared between the two ant species. Additionally, mass spectrometry metabolomics analysis of metabolites on the cuticle of ants, which excludes the detection of cuticular hydrocarbons produced by the host, were conducted to evaluate correlations among bacterial OTUs and m/z ion mass. Although some positive and negative correlations are found, the cuticular chemical composition was weakly species-specific, suggesting that cuticular bacterial communities are prominently environmentally acquired. Overall, our results suggest the environment is the dominant source of bacteria found on the cuticle of ants. © 2020 John Wiley & Sons Ltd

  Abstract High-throughput sequencing of the 16S rRNA gene has considerably helped revealing the essential role of bacteria living on insect cuticles in the ecophysiology and behaviour of their hosts. However, our understanding of host-cuticular microbiota feedbacks remains hampered by the difficulties of working with low bacterial DNA quantities as with individual insect cuticle samples, which are more prone to molecular biases and contaminations. Herein, we conducted a methodological benchmark on the cuticular bacterial loads retrieved from two Neotropical ant species of different body size and ecology: Atta cephalotes (~15 mm) and Pseudomyrmex penetrator (~5 mm). We evaluated the richness and composition of the cuticular microbiota, as well as the amount of biases and contamination produced by four DNA extraction protocols. We also addressed how bacterial community characteristics would be affected by the number of individuals or individual body size used for DNA extraction. Most extraction methods yielded similar results in terms of bacterial diversity and composition for A. cephalotes (~15 mm). In contrast, greater amounts of artefactual sequences and contaminations, as well as noticeable differences in bacterial community characteristics were observed between extraction methods for P. penetrator (~5 mm). We also found that large (~15 mm) and small (~5 mm) A. cephalotes individuals harbour different bacterial communities. Our benchmark suggests that cuticular microbiota of single individual insects can be reliably retrieved provided that blank controls, appropriate data cleaning, and individual body size and functional role within insect society are considered in the experiment.

  Abstract Light is of primary importance in structuring tropical tree communities. Light exposure at seedling and adult stages has been used to characterize the ecological profile of tropical trees, with many implications in forest management and restoration ecology. Most shade-tolerance classification systems have been proposed based on empirical observations in a specific area and thus result in contradictions among categories assigned to a given species. In this study, we aimed to quantify the light requirements for seedling growth of a Central African timber tree, Lophira alata (Ochnaceae), taking into account effects of population origin. In two controlled experiments: a light response experiment and a comparative population experiment, conducted in southwestern Cameroon, using seeds collected from four populations (three from Cameroon and one from Gabon), we examined the quantitative responses to irradiance of seedlings. After 2 years, mortality was very low (<3%), even in extremely low irradiance. Growth and biomass allocation patterns varied in response to light, with intermediate irradiance (24–43%) providing optimal conditions. Light response differed between populations. The Boumba population in the northeastern edge of the species' distribution exhibited the highest light requirements, suggesting a local adaptation. As a result of positive growth at low irradiance and maximum growth at intermediate irradiance, we concluded that L. alata exhibits characteristics of both non-pioneer and pioneer species. Implications of our results to propose an objective way to assign the light requirement for tropical tree species are discussed.

  Abstract The expansion of selective logging in tropical forests may be an important source of global carbon emissions. However, the effects of logging practices on the carbon cycle have never been quantified over long periods of time. We followed the fate of more than 60 000 tropical trees over 23 years to assess changes in aboveground carbon stocks in 48 1.56-ha plots in French Guiana that represent a gradient of timber harvest intensities, with and without intensive timber stand improvement (TSI) treatments to stimulate timber tree growth. Conventional selective logging led to emissions equivalent to more than a third of aboveground carbon stocks in plots without TSI (85 Mg C/ha), while plots with TSI lost more than one-half of aboveground carbon stocks (142 Mg C/ha). Within 20 years of logging, plots without TSI sequestered aboveground carbon equivalent to more than 80% of aboveground carbon lost to logging (-70.7 Mg C/ha), and our simulations predicted an equilibrium aboveground carbon balance within 45 years of logging. In contrast, plots with intensive TSI are predicted to require more than 100 years to sequester aboveground carbon lost to emissions. These results indicate that in some tropical forests aboveground carbon storage can be recovered within half a century after conventional logging at moderate harvest intensities.
