Abstract |
Many tropical tree species generate natural decay resistance by producing bioactive metabolites. Among them, Sextonia rubra (Lauraceae) is a widely exploited species for construction in French Guiana. Rubrynolide and rubrenolide, which are secondary metabolites isolated from the stem wood of S. rubra, exhibit potent antifungal and termiticidal activities that result in the exceptional durability of the heartwood. Although the structure and total synthesis of these two biologically active compounds have been well described in literature, little is known about their cellular localization or biosynthesis process in living trees. Time-of-flight secondary ion mass spectrometry (TOF-SIMS) has been well known for its high lateral resolution imaging with a typical beam size of 0.4-2 µm, thus sufficient to image the wood surface at subcellular level.
Wood surfaces of sapwood, transition zone and heartwood from living trees of S. rubra were prepared with ultramicrotome using diamond knife. Each surface was mapped by Bi3+ cluster TOF-SIMS with high lateral resolution (~ 0.4 µm) which was achieved by employing burst alignment primary ion focusing mode with an extraction delay of the secondary ions to improve mass resolution. 3D imaging of a multi-structure wood sample was performed using dual beam depth profiling method, with 25 keV Bi3+ and 10 keV Ar1000+ as analysis and sputter beam, respectively. To examine the effect of life-history traits on the production of bioactive metabolites, the corresponding counterparts from a tree which had suffered stress during growth were investigated as well. |

Abstract |
L’incidence des infections fongiques a augmenté ces dernières années en raison d’un nombre croissant de patients à hauts risques. Ce fait, conjugué à la résistance aux antibiotiques et à la toxicité rencontrée sur le long terme pour certains traitements, a conduit à de nombreuses recherches sur de nouveaux traitements des mycoses. En conséquence, il existe un intérêt croissant pour les plantes médicinales comme alternatives aux médicaments synthétiques. Les huiles essentielles sont des mélanges complexes de composés volatiles isolés par hydrodistillation. Ces volatiles sont pour la plupart des terpénoïdes (mono- et sesquiterpènes). L’objectif de cette étude est d’identifier de nouvelles huiles essentielles présentant des activités antifongiques et de déterminer leur composition chimique. Les huiles essentielles étudiées ici n’ont pas été choisies en fonction d’un usage traditionnel mais ont été extraites à partir de plantes très présentes en Guyane Française. La composition chimique des huiles de fruits de Protium heptaphyllum et des parties aériennes de Otacanthus caeruleus ont été déterminées par CPG/SM et par analyse RMN, avant et après fractionnement (en chromatographie sur colonne de gel de silice) et l’activité antifongique des huiles a été étudiée contre plusieurs champignons ou levures (Microsporum canis, Microsporum gypseum, Trichophyton mentagrophytes, Candida krusei, Candida parapsilosis). |

Abstract |
En Guyane, la conversion de sols forestiers en sols agricoles engendre de multiples conséquences sur le milieu ; en particulier, le bilan des flux de gaz à effet de serre est modifié. Notre étude se base sur une approche comparative entre quatre occupations du sol : parcelles cultivées en semis direct, parcelles cultivées en semis conventionnel, la prairie et la situation de référence, la forêt. La méthode des chambres statiques au champ a permis de mesurer sur une année, les flux de N2O et de CO2. Des mesures concernant l’azote minéral, l’humidité et la température du sol, ont été réalisées. La compréhension de ces déterminants des flux permet d’évaluer l’impact des pratiques culturales sur l’activité des microorganismes du sol, principaux responsables des émissions de GES par les sols via la minéralisation, la nitrification et la dénitrification. Les résultats ont montré que le semis direct est la pratique culturale qui stocke le plus de carbone (11,09 ±3,41 tC.ha-1.an-1) et dont les émissions de CO2 sont les plus faibles (29,98±2,58 kg C.ha-1.jour-1). Cependant, le semis direct est également une pratique émettrice de grandes quantités de N2O (7,52±2,75 g N.ha-1.jour-1). |

L’UMR Ecofog étudie les forêts tropicales guyanaises, leur diversité et leur fonctionnement L’équipe « Ecologie des Communautés et des Ecosystèmes » comprenant le laboratoire de microbiologie des sols a pour mission d’étudier certains organismes pour mieux les comprendre. Dans les sols guyanais d’une part des bactéries sont capables de nitrifier et de dénitrifier en anaérobiose mais leur identification n’est pas encore faite, d’autre part il existe un panel de champignons qui ne sont pas répertoriés. Par conséquent il faut identifier les bactéries capables de nitrifier et dénitrifier en anaérobiose mais aussi lister les champignons récoltés sur divers sites. Cette identification se fait à l’aide des outils de biologie moléculaire qui permettent d’obtenir des séquences à partir de l’ADN extrait des souches bactériennes isolées en culture ou de fragments de champignons récoltés. Les séquences obtenues sont confrontées aux bases de données de référence ce qui permet d’identifier les espèces en présence. Ce stage de six semaines a mis en évidence 7 souches bactériennes anaérobies isolées des sols et devrais permettre de cataloguer des champignons. |
